FAQ Genome Sequencing

Re-Sequencing of Exons & Genome Fragments

Re-Sequencing is the sequencing of pre-sequenced and pre-selected genomic regions such as exons in order to find a correlation between sequence deviations, for instance SNPs or deletions and phenotypes.

Eurofins MWG Operon offers a full service in 3 phases from (I) primer design, primer synthesis and establishment of PCR conditions, (II) to high-throughput PCR and double strand sequencing concluding with bioinformatic analysis, (III) to determine differences between reference and samples.

Specifications

Phase I: Establishment of the PCR

  • Primer design for all exon regions
  • Primer synthesis in HPSF® quality
  • Establishment of PCR amplification on two DNA samples
  • Double strand sequencing of the PCR fragments
  • Two repetitions of PCR setup in the event of failure

Phase II: PCR and Sequencing

  • High throughput PCR of all samples
  • Purification of PCR products
  • High throughput sequencing of PCR products
  • One repetition in the event of failure

Phase III: Bioinformatic Analysis

  • Double Strand sequencing recommended
  • Identification of all differences between reference and samples
  • Homozygous substitutions
  • Heterozygous substitutions
  • Detection of insertions/deletions
  • Mutation report and comparison of all samples provided.
