Excellent sequencing service of pro- and eukaryotic genomes of any size - from shotgun coverage sequencing to complete finishing and data analysis including gap closure and editing.
High quality genome assemblies are obtained by sequencing a combination of shotgun libraries and long-paired end (LPE)-libraries on the Roche GS FLX.
The LPE fragments comprise two DNA sequence runs bridged by 3 kbp, 8 kbp, 20 kbp or 40 kbp genome regions (jumping distance). This unique 4 library approach has proven to result in the longest average scaffold sizes and smallest number of remaining gaps.
Proven experience in prokaryotic and eukaryotic high quality de novo genome sequencing.
In close cooperation with our customers, we perform de novo genome sequencing projects in the following different stages:
- Ultra high throughput sequencing of a shotgun library and assembly into contigs
- Scaffolding of contigs by long paired end sequencing
- Closing of gaps based on Sanger technology
- Bioinformatic analysis, e.g. annotation or comparison of different organisms
Depending on the aim of your studies, resequencing projects can be addressed using either Illumina HiSeq 2000 or Roche GS FLX technology:
- Affordable resequencing with Illumina HiSeq 2000
For SNPdetection and comparative analysis to a reference genome, we recommend the cost-efficient resequencing setup with Illumina HiSeq 2000 technology. - Sophisticated projects with Roche GS FLX
If large indels or genomic correlations between the sequenced and the reference genome are likely to be detected, sequencing on Roche GS FLX may be the technology of choice. The longer reads of Roche GS FLX facilitate de novo assembly and subsequent comparative analysis. With this approach, you benefit from high quality genome sequences even if large indels or genomic arrangements occur.
The Roche GS Junior is the optimum system for the sequencing of smaller scale projects (e.g. BACs) which require only one eighth of the capacity of a GS FLX Titanium run or which would not be suitable for an Illumina run. As a consequence, the GS Junior adds a higher degree of flexibility in project planning and can have a positive impact on the turnaround time for small sized projects.
Depending on your final objective, we will assist you in designing the appropriate sequencing experiment for your research. In close consultation with you, we will work out the strategy to deliver the best possible data quality.
