Custom DNA & RNA Synthesis

Every oligonucleotide passes strict quality controls before it is delivered to our customers. Through continuous process improvements over the years and investigations into advanced state-of-the-art automation systems for synthesis, cleavage, purification and quality control instruments, Eurofins MWG Operon has become one of the world’s leading providers for custom DNA and RNA oligonucleotides.

Manufacturing Process

1. Online Ordering
Your online order reaches Eurofins MWG Operon via our online ordering system (Ecom) and enters our completely barcode driven LIMS system, which monitors your oligonucleotide through the whole production process.

2. Synthesis
Your oligonucleotides are produced by using phosphoramidite chemistry on our proprietary Eurofins MWG Operon synthesiser technology. Constant in-process-monitoring of the trityl values ensures the quality of the synthesis

3. Cleavage
The synthesised oligonucleotides are then cleaved from the synthesis matrix using a highly efficient chemical method.

4. Purification
The crude oligonucleotides are then purified, according to the type of oligonucleotide and purification option you have chosen (HPSF®, RP-HPLC or HYPUR®)

5. Measurement of Optical Density (OD)
We subsequently quantify all oligonucleotides by measuring the optical density (OD), using a highly parallel process. The OD values are transferred into our LIMS system automatically

6. Quality End Control
For quality control, each and every one of your oligonucleotides is checked by analytical MALDI-TOF mass spectroscopy, using the latest state-of-the-art technology. Additional analysis by capillary gel electrophoresis (CGE) is performed on unmodified oligonucleotides, which have been purified by RP-HPLC or HYPUR®

7. Normalisation and Delivery
As a last step, the oligonucleotides are then formatted to your specifications. Unmodified oligonucleotides in tube format are lyophilised by default. Standard normalisations are available in tubes, 96- or 384well plates. They can be adjusted to 50 – 500 pmol/µl; forward and reverse primers can be combined in “Mixture Plates”. Aliquoting options are also available

8. Tracking and Order History
Whenever you order via our online Ecom system, you can track the current production and delivery status of your oligonucleotides at any time. Additionally you can re-order specific oligonucleotides from your order history within one year.
